Principle:
TG concentration is measured by a timed endpoint method. The Synchron® System automatically proportions the appropriate sample and reagent volumes into the cuvette at a ratio of 1:100. Triglycerides in the sample are hydrolyzed to glycerol and free fatty acids by the action of lipase. A sequence of three coupled enzymatic steps using glycerol kinase, glycerophosphate oxidase and horseradish peroxidase cause the oxidative coupling of 3,5-dichloro2-hydroxybenzenesulfonic acid with 4-aminoantipyrine to form a red quinoneimine dye. The change in absorbance at 520 nm is directly proportional TG concentration in the sample, and is used by the system to calculate and report TG concentration.
